Study of DMC 1 gene expression encoding meiotic recombination enzyme during different stages of bud formation in Bell pepper (Capsicum annuum L.)

Abstract :
The bell pepper (Capsicum annuum) of the Capsicum genus and of the Solanaceae family. This plant is native to Central America and Mexico. The bell pepper is a full-flowered diploid plant (2n=2x=24). The meiotic recombination occurs through a crossover between the homologues pair of the mother and fathers chromosomes; in this case, a set of chromosomes are randomly placed together. Biochemical and structural studies show that the two DMC1 and RAD51 enzymes have similarities in DNA pairing and strand exchange activities in meiotic crossover. At DMC1 enzyme activity sites, crossover occurs more often. This study was conducted to evaluate the expression of DMC 1 gene in growth stages, including 10-leaf stage, budding start, buds size Less than of 2.5 mm (pre mitotic phase) and buds size of 2.5-4.5 mm (tetrad formation stage) and buds of size 8-11 mm (mature pollen). Measurement of the relative expression of DMC 1 gene was investigated using the SYBR Green Master Mix (BioFACT) and relative to the GAPDH reference gene in the ABI StepOnePlus Real-Time PCR device. The results of analysis of variance data indicated that there was a significant difference between growth stages in the expression of DMC 1 gene at a probability level of 1. Compare Mean showed that the pre mitotic phase treatment had the highest relative expression of DMC 1 gene and the tetrad formation stage treatment was in the next position. 10-leaf stage, mature pollen and budding start treatments, had also the lowest relative expression of DMC 1 gene, respectively.
